There has been so much elucidated and examined as a result of observations made as immune checkpoint inhibitors began to be used clinically. I’m particularly struck by the research into the intestinal biome and its interactions with the immune system. Ipilimumab (the prototypical CTLA4blocker) has a black box warning because of the risk of severe immune-mediated toxicities, including enterocolitis, the risk of which is tied to the types of intestinal bacteria present and also to the response in treating melanoma. It is oddly beautiful.

I worked in a tumor immunology lab briefly using anti-ctla4 in liver cancer, this was such a fun read, takes me back!!
